Flow cytometric analysis of human lymphocytes stained using a selection of mouse primary antibodies against human targets, detected using mouse IgG2b specific Ankyron clone CS30121 (0.1 μg Ankyron, in red). Negative control of Ankyron alone (no primary antibody) in grey. The Ankyron selectively binds to the anti-human CD8a mouse IgG2b primary antibody positive control, with no specific signal observed for the negative control mouse IgG1, IgG2a, IgG3 and IgM primary antibodies.
Binding assay determination of the signal to noise ratio (SNR) of V5-labeled Ankyrons® CS30121 binding to 0.5 picomoles of immobilised IGG2B_MOUSE, over a background (BG) of non-cognate target protein. The Ankyron was pre-conjugated with the ProImmune Anti-V5 HRP label reagent before detection with a chemiluminescence substrate.
Affinity measurement of V5-labeled Ankyrons® CS30121 binding to IGG2B_MOUSE, of value Kd = 1.58 nM, with 95% confidence bounds (CI95U, CI95L). Conjugation to ProImmune Anti-V5 HRP label reagent allows detection in Solution Equilibrium Titration immunoassay with a chemiluminescent substrate.
