ProVE SL – Self-loading MHC Class I Monomers
ProVE® SL Self-loading MHC Class I Monomers
maximum flexibility epitope exploration across an extended allele range

Stable MHC Class I Monomers for you to add your own peptide
ProVE SL are novel empty biotinylated MHC Class I Monomers with superior stability that allow rapid self-loading with peptides for T cell detection. Unlike monomers that require peptide exchange peptides can simply be added to load empty complexes.
The most flexible way to produce customized MHC tetramers for T cell detection
-
- Use peptides you already have in your lab
-
- Plan and execute and re-configure experiments quickly
-
- Extended range of alleles allows you to get the most out of your existing donor cohort
-
- Work flexibly with any avidin / streptavidin conjugate available, including fluorescent, barcode and mass labels
-
- Can be readily immobilized on streptavidin-coated beads or surfaces

Figure 2: Schematic overview on how to use your ProVE SL Self-loading MHC Class I Monomers. Once you receive your empty biotinylated ProVE SL Monomers, simply incubate with your selected peptide for 24 hours to load. Then, the loaded monomers may be conjugated to fluorophore-labelled streptavidin to tetramerize. The resulting pMHC tetramers can be used for specific detection of antigen-specific CD8+ T cells by flow cytometry.
Features of ProVE SL MHC Class I Monomers

-
- Long-term storage
ProVE SL Staining for Antigen-Specific CD8+ T cells

Figure 3. Comparison staining between classical and ProVE SL tetramers (A) CMV-specific CD8+ T cells stained using HLA-A*02:01 / NLVPMVATV fully pre-synthesized MHC Class I tetramer. (B) Independent staining of CMV-specific CD8+ T cells using A*02:01-restricted ProVE SL monomers, loaded with NLVPMVATV peptide and tetramerized with streptavidin. (C) Independent staining of CMV-specific CD8+ T cells using A*02:01-restricted ProVE SL monomers, loaded with irrelevant non-binding negative control peptide and tetramerized with streptavidin. (D) Staining of CMV-specific CD8+ T cells using B*07:02-restricted ProVE SL monomers, loaded with TPRVTGGGAM peptide and tetramerized with streptavidin. (E) Staining of CMV-specific CD8+ T cells using B*07:02-restricted ProVE SL monomers, loaded with irrelevant non-binding negative control peptide and tetramerized with streptavidin. Please note that results may vary based on peptide quality and affinity.
More detail on how using ProVE SL Monomers can help you
- Study donor cohorts for antigen specific immune responses according to their actual tissue types found. Here you are not limited by the most common alleles, but you can potentially cover all A B C locus alleles present in particular donors
- Make the most of donors cells you have actually banked taking into account more MHC alleles
- Carry out population wide studies to understand antigen specific immune responses
- Explore antigen specific immune responses of particular interest in people with less common alleles, such as protection linked alleles or rapid progressor linked alleles in a particular response
- Carry out tetramer guided epitope mapping, including with multi-colour flow panels or mass cytometry by conjugating a range of streptavidin fluorochrome conjugates
- Configure your own reagents quickly and flexibly, according to your evolving interests;compatible with barcoding, mass cytometry and other advanced methods
- Need peptides for your study? Our thinkpeptides product range offers you a full range of immunology peptide solutions including peptides for self-loading
- Don’t know the full HLA type of your donors? Find out the full detail with our convenient world wide typeHLA tissue typing service
- Connect with our market leading applications science team for help with structuring your project
- Can be easily combined with single cell RNA seq kits
