Ankyrons® Testing Data
HUVEC were incubated with Cobalt (II) chloride hexahydrate overnight (to stabilise hypoxia inducible factors), then fixed in formaldehyde (4%) and permeabilised with 0.5% Triton X-100. Cells were stained using 1.4 µg V5-tagged Ankyron and anti-V5 antibody conjugated with APC (red) in Permeabilisation Staining Buffer (RPMI 1640 + 10 % human serum + 0.5 % saponin), counterstaining nuclei with DAPI (blue) and F-actin with Phalloidin. Binding assay determination of the signal to noise ratio (SNR) of V5-labeled Ankyrons® AW40110 binding to 0.5 picomoles of immobilised HIF1A_HUMAN, over a background (BG) of non-cognate target protein. The Ankyron was pre-conjugated with the ProImmune Anti-V5 HRP label reagent before detection with a chemiluminescence substrate.
Flow cytometric analysis of CD8+ T-cells (CD3+CD8+) and NK cells (CD3-NKp46+) from BALB/c kaede mice, following subcutaneous injections with CT26 murine colorectal carcinoma cells. Cells were fixed in BD Cytofix buffer, permeabilized with FoxP3 buffer and stained overnight at room temperature with 3 µg ANK00655_1_AW40110 per well, detecting with HisTag-AF647. Negative control: Fluorescence minus one, FMO, with no Ankyron present.
