Confocal microscopy of Aag2-AF2 Aedes aegypti cells, either treated with double-stranded (ds) RNA (mock), dsGFP, dsRel1A (which is processed by insect cells into siRNAs to silence Rel1A expression, or untreated. Cells were stained with V5-tagged Ankyron, then mouse anti-V5 antibody followed by fluorescent anti-mouse antibody for detection (grey scale), with nuclei counterstained with DAPI (blue). Binding assay determination of the signal to noise ratio (SNR) of V5-labeled Ankyrons® BT40163 binding to 0.5 picomoles of immobilised Q171A6_AEDAE, over a background (BG) of non-cognate target protein. The Ankyron was pre-conjugated with the ProImmune Anti-V5 HRP label reagent before detection with a chemiluminescence substrate.

Affinity measurement of V5-labeled Ankyrons® BT40163 binding to Q171A6_AEDAE, of value K_d = 198.0 nM, with 95% confidence bounds (CI95_U, CI95_L). Conjugation to ProImmune Anti-V5 HRP label reagent allows detection in Solution Equilibrium Titration immunoassay with a chemiluminescent substrate.
