Ankyrons® Testing Data
Mouse macrophage J774A.1 cells were cultured overnight in either the absence or presence of LPS, then stained using 1.4 µg V5-tagged Ankyron and anti-V5 antibody conjugated with APC (red) in Staining Buffer (RPMI 1640 + 10 % human serum), counterstaining nuclei with DAPI (blue). Flow cytometric analysis of mouse J774A.1 cells, either resting (blue) or following stimulation (red) overnight with LPS, prior to staining with pre-conjugated Ankyron (0.4 µg) and anti-V5 APC. Negative controls, anti-V5 APC only on unstimulated (light grey) and stimulated (dark grey) cells.
Binding assay determination of the signal to noise ratio (SNR) of V5-labeled Ankyrons® AO40285 binding to 0.5 picomoles of immobilised CD80_MOUSE, over a background (BG) of non-cognate target protein. The Ankyron was pre-conjugated with the ProImmune Anti-V5 HRP label reagent before detection with a chemiluminescence substrate.
Affinity measurement of V5-labeled Ankyrons® AO40285 binding to CD80_MOUSE, of value Kd = 106.0 nM, with 95% confidence bounds (CI95U, CI95L). Conjugation to ProImmune Anti-V5 HRP label reagent allows detection in Solution Equilibrium Titration immunoassay with a chemiluminescent substrate.
