Binding assay determination of the signal to noise ratio (SNR) of V5-labeled Ankyrons® AO40203 binding to 0.5 picomoles of immobilised CD7_MOUSE, over a background (BG) of non-cognate target protein. The Ankyron was pre-conjugated with the ProImmune Anti-V5 HRP label reagent before detection with a chemiluminescence substrate.
Flow cytometric analysis of CD8+ T-cells (CD3+CD8+) from the liver of C57BL6/J mice, 30 days post infection with Lymphocytic choriomeningitis virus Clone 13. Cells were stained with preconjugated Ankyron (1µg) and anti-V5 PE (preconjugated at 4ºC for 15 minutes), 30 minutes at 4ºC. Cells were then washed before staining for other surface antibodies and run unfixed on a 5 laser Cytek Aurora. Negative control: Fluorescence minus one, FMO, with no primary or secondary Ankyron present (blue), and a no-primary control, with anti-V5 Ankyron PE alone (red). Flow cytometric analysis of congenically distinct endogenous CD8+ T cells (C57BL/6J, CD45.2+, grey) and either CD7 wild-type (WT, CD45.1+ CD45.2-, blue) or CD7 knockout (KO, CD45.1+CD45.2+, red) cells in mouse spleen following infection with Lymphocytic choriomeningitis virus LCMV). Staining was performed using V5-tagged Ankyron, a V5-specific nanobody recombinantly expressed as rabbit IgG-Fc fusion protein, and polyclonal anti-rabbit IgG antibodies conjugated with AlexaFluor 647.
