Ankyrons® Testing Data

Human monocytes were fixed in formaldehyde (4%), then stained using 5 µg V5-tagged Ankyron and anti-V5 antibody conjugated with APC (red) in Staining Buffer (RPMI 1640 + 10 % human serum), counterstaining nuclei with DAPI (blue). Image captured by epifluorescence microscopy.
Flow cytometric analysis of human monocytes, following incubation with 2.5 μg Ankyron and detecting with anti-V5-APC, (red). Negative control: anti-V5-APC alone, with no Ankyron present (grey).
Binding assay determination of the signal to noise ratio (SNR) of V5-labeled Ankyrons® AB30169 binding to 0.5 picomoles of immobilised CD14_HUMAN, over a background (BG) of non-cognate target protein. The Ankyron was pre-conjugated with the ProImmune Anti-V5 HRP label reagent before detection with a chemiluminescence substrate.
Affinity measurement of V5-labeled Ankyrons® AB30169 binding to CD14_HUMAN, of value Kd = 0.0976 nM, with 95% confidence bounds (CI95U, CI95L). Conjugation to ProImmune Anti-V5 HRP label reagent allows detection in Solution Equilibrium Titration immunoassay with a chemiluminescent substrate.
